Uncertain significance for Atrial fibrillation, familial, 7 — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_002234.4(KCNA5):c.230C>G (p.Pro77Arg), citing Invitae Variant Classification Sherloc (09022015): This variant is present in population databases (no rsID available, gnomAD 0.002%). This sequence change replaces proline, which is neutral and non-polar, with arginine, which is basic and polar, at codon 77 of the KCNA5 protein (p.Pro77Arg). This variant has not been reported in the literature in individuals affected with KCNA5-related conditions. ClinVar contains an entry for this variant (Variation ID: 653091). Algorithms developed to predict the effect of missense changes on protein structure and function are either unavailable or do not agree on the potential impact of this missense change (SIFT: "Deleterious"; PolyPhen-2: "Benign"; Align-GVGD: "Class C0"). In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
