Uncertain significance for Charcot-Marie-Tooth disease dominant intermediate E; Focal segmental glomerulosclerosis 5 — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_022489.4(INF2):c.1587dup (p.Val530fs), citing Invitae Variant Classification Sherloc (09022015): This sequence change creates a premature translational stop signal (p.Val530Argfs*50) in the INF2 gene. It is expected to result in an absent or disrupted protein product. However, the current clinical and genetic evidence is not sufficient to establish whether loss-of-function variants in INF2 cause disease. The frequency data for this variant in the population databases is considered unreliable, as metrics indicate poor data quality at this position in the gnomAD database.
