Loss-of-function variants in NF2 are known to be pathogenic (PMID: 9643284, 16983642). This variant has been observed in an individual with clinical features of neurofibromatosis type 2 (PMID:¬†18033041). This variant is an out-of-frame deletion of the genomic region encompassing exon 12 of the NF2 gene. This is expected to create a premature translational stop signal and result in an absent or disrupted protein product.
