Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_005732.4(RAD50):c.211A>G (p.Lys71Glu), citing Ambry Variant Classification Scheme 2023: The p.K71E variant (also known as c.211A>G), located in coding exon 2 of the RAD50 gene, results from an A to G substitution at nucleotide position 211. The lysine at codon 71 is replaced by glutamic acid, an amino acid with similar properties. This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
