Uncertain significance for Autosomal recessive limb-girdle muscular dystrophy type 2W — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_001161403.3(LIMS2):c.12-6783G>A, citing Invitae Variant Classification Sherloc (09022015): While this variant is not present in population databases, the frequency information is unreliable, as metrics indicate poor data quality at this position in the ExAC database. This sequence change replaces arginine with glutamine at codon 15 of the LIMS2 protein (p.Arg15Gln). The arginine residue is weakly conserved and there is a small physicochemical difference between arginine and glutamine. This variant has not been reported in the literature in individuals with LIMS2-related disease. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance. Algorithms developed to predict the effect of sequence changes on RNA splicing suggest that this variant may create or strengthen a splice site, but this prediction has not been confirmed by published transcriptional studies.
