This variant lies in the MCCC2 gene (transcript NM_022132.5) at coding-DNA position 506, where A is replaced by G; at the protein level this means replaces tyrosine at residue 169 with cysteine — a missense variant. Submitter rationale: This sequence change replaces tyrosine, which is neutral and polar, with cysteine, which is neutral and slightly polar, at codon 169 of the MCCC2 protein (p.Tyr169Cys). This variant is not present in population databases (gnomAD no frequency). This missense change has been observed in individual(s) with 3-methylcrotonyl-CoA carboxylase deficiency (internal data). In at least one individual the data is consistent with being in trans (on the opposite chromosome) from a pathogenic variant. It has also been observed to segregate with disease in related individuals. ClinVar contains an entry for this variant (Variation ID: 652543).
