Uncertain significance for Hereditary sensory and autonomic neuropathy type 7; Familial episodic pain syndrome with predominantly lower limb involvement — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NC_000003.12:g.(?_38925395)_(38925529_?)del, citing Invitae Variant Classification Sherloc (09022015): This variant is an out-of-frame deletion of the genomic region encompassing exon 5 of the SCN11A gene. This is expected to create a premature translational stop signal and result in an absent or disrupted protein product. This variant has not been reported in the literature in individuals with SCN11A-related disease. The current clinical and genetic evidence is not sufficient to establish whether loss-of-function variants in SCN11A cause disease. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
