Pathogenic for Pyridoxine-dependent epilepsy — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_001182.5(ALDH7A1):c.1003C>T (p.Arg335Ter), citing Invitae Variant Classification Sherloc (09022015): This sequence change creates a premature translational stop signal (p.Arg335*) in the ALDH7A1 gene. It is expected to result in an absent or disrupted protein product. Loss-of-function variants in ALDH7A1 are known to be pathogenic (PMID: 16491085, 20554659). This variant is present in population databases (no rsID available, gnomAD 0.01%). This premature translational stop signal has been observed in individual(s) with pyridoxine-dependent epilepsy (PMID: 17068770, 23350806, 24664088). In at least one individual the data is consistent with being in trans (on the opposite chromosome) from a pathogenic variant. This variant is also known as c.919C>T (p.Arg307X).
