NM_000138.5(FBN1):c.5639T>C (p.Phe1880Ser) was classified as Uncertain significance for Marfan syndrome; Familial thoracic aortic aneurysm and aortic dissection by Labcorp Genetics (formerly Invitae), Labcorp, citing Invitae Variant Classification Sherloc (09022015): In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance. Algorithms developed to predict the effect of missense changes on protein structure and function are either unavailable or do not agree on the potential impact of this missense change (SIFT: "Deleterious"; PolyPhen-2: "Probably Damaging"; Align-GVGD: "Class C0"). This variant has been observed in an individual affected with thoracic aortic aneurysm and dissection (Invitae). This variant is not present in population databases (ExAC no frequency). This sequence change replaces phenylalanine with serine at codon 1880 of the FBN1 protein (p.Phe1880Ser). The phenylalanine residue is highly conserved and there is a large physicochemical difference between phenylalanine and serine.
