Uncertain significance for Myopathy, areflexia, respiratory distress, and dysphagia, early-onset — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_001256545.2(MEGF10):c.2570G>C (p.Gly857Ala), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces glycine with alanine at codon 857 of the MEGF10 protein (p.Gly857Ala). The glycine residue is highly conserved and there is a small physicochemical difference between glycine and alanine. This variant is not present in population databases (ExAC no frequency). This variant has not been reported in the literature in individuals with MEGF10-related disease. Algorithms developed to predict the effect of missense changes on protein structure and function output the following: SIFT: "Deleterious"; PolyPhen-2: "Benign"; Align-GVGD: "Class C0". The alanine amino acid residue is found in multiple mammalian species, suggesting that this missense change does not adversely affect protein function.
