NC_000008.11:g.(?_99661334)_(99661511_?)del was classified as Pathogenic for Cohen syndrome by Labcorp Genetics (formerly Invitae), Labcorp, citing Invitae Variant Classification Sherloc (09022015): A similar copy number variant has been observed in individual(s) with VPS13B-related disease (Invitae). In at least one individual the data is consistent with being in trans (on the opposite chromosome) from a pathogenic variant. It has also been observed to segregate with disease in related individuals. Experimental studies and prediction algorithms are not available or were not evaluated, and the functional significance of this variant is currently unknown. For these reasons, this variant has been classified as Pathogenic. This variant is a gross deletion of the genomic region encompassing exon(s) 35 of the VPS13B gene. This variant would be expected to be in-frame, preserving the integrity of the reading frame.
