Pathogenic for Neuronal ceroid lipofuscinosis — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NC_000015.10:g.(?_68229482)_(68229604_?)del, citing Invitae Variant Classification Sherloc (09022015): This variant is a gross deletion of the genomic region encompassing exon 1 of the CLN6 gene, which includes the initiator codon. The 5' end of this event is unknown as it extends beyond the assayed region for this gene and therefore may encompass additional genes. The 3' boundary is likely confined to intron 1 of the CLN6 gene. This is expected to result in an absent or disrupted protein product. A similar deletion of exon 1 has been reported in an individual affected with a CLN6-related condition (PMID: 26115733). Loss-of-function variants in CLN6 are known to be pathogenic (PMID: 19135028). For these reasons, this variant has been classified as Pathogenic.
