Pathogenic for CHARGE syndrome — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_017780.4(CHD7):c.2700del (p.Pro900_Val901insTer), citing Invitae Variant Classification Sherloc (09022015). This variant lies in the CHD7 gene (transcript NM_017780.4) at coding-DNA position 2700, deleting one base. Submitter rationale: For these reasons, this variant has been classified as Pathogenic. Loss-of-function variants in CHD7 are known to be pathogenic (PMID: 22461308, 25077900). Algorithms developed to predict the effect of sequence changes on RNA splicing suggest that this variant may create or strengthen a splice site, but this prediction has not been confirmed by published transcriptional studies. This variant has not been reported in the literature in individuals with CHD7-related disease. This variant is not present in population databases (ExAC no frequency). This sequence change creates a premature translational stop signal (p.Val901*) in the CHD7 gene. It is expected to result in an absent or disrupted protein product.
