Uncertain significance for Atelosteogenesis type II; Multiple epiphyseal dysplasia type 4; Achondrogenesis, type IB; Diastrophic dysplasia — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_000112.4(SLC26A2):c.892C>A (p.His298Asn), citing Invitae Variant Classification Sherloc (09022015). This variant lies in the SLC26A2 gene (transcript NM_000112.4) at coding-DNA position 892, where C is replaced by A; at the protein level this means replaces histidine at residue 298 with asparagine — a missense variant. Submitter rationale: This sequence change replaces histidine with asparagine at codon 298 of the SLC26A2 protein (p.His298Asn). The histidine residue is moderately conserved and there is a small physicochemical difference between histidine and asparagine. This variant is not present in population databases (ExAC no frequency). This variant has not been reported in the literature in individuals affected with SLC26A2-related conditions. Algorithms developed to predict the effect of missense changes on protein structure and function (SIFT, PolyPhen-2, Align-GVGD) all suggest that this variant is likely to be tolerated. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
