Uncertain significance for Primary ciliary dyskinesia 32 — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_031924.8(RSPH3):c.-414C>G, citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces proline with alanine at codon 5 of the RSPH3 protein (p.Pro5Ala). The proline residue is weakly conserved and there is a small physicochemical difference between proline and alanine. This variant is present in population databases (rs747323201, ExAC 0.003%). This variant has not been reported in the literature in individuals affected with RSPH3-related conditions. Algorithms developed to predict the effect of missense changes on protein structure and function output the following: SIFT: "Tolerated"; PolyPhen-2: "Benign"; Align-GVGD: "Class C0". The alanine amino acid residue is found in multiple mammalian species, which suggests that this missense change does not adversely affect protein function. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
