Loss-of-function variants in NF1 are known to be pathogenic (PMID: 10712197, 23913538). A different variant (c.7901delC) giving rise to the same protein effect observed here (p.Pro2634Leufs*24) has been determined to be pathogenic (PMID: 15146469). This suggests that this variant is also likely to be causative of disease. This variant has not been reported in the literature in individuals with NF1-related disease. This variant is not present in population databases (ExAC no frequency). This sequence change creates a premature translational stop signal (p.Pro2634Leufs*24) in the NF1 gene. It is expected to result in an absent or disrupted protein product.
