Uncertain significance — the classification assigned by Ambry Genetics to NM_021930.6(RINT1):c.1621A>G (p.Asn541Asp), citing Ambry Variant Classification Scheme 2023: The p.N541D variant (also known as c.1621A>G), located in coding exon 11 of the RINT1 gene, results from an A to G substitution at nucleotide position 1621. The asparagine at codon 541 is replaced by aspartic acid, an amino acid with highly similar properties. This amino acid position is highly conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
