Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_006231.4(POLE):c.2812C>T (p.Leu938Phe), citing Ambry Variant Classification Scheme 2023: The p.L938F variant (also known as c.2812C>T), located in coding exon 24 of the POLE gene, results from a C to T substitution at nucleotide position 2812. The leucine at codon 938 is replaced by phenylalanine, an amino acid with highly similar properties. This amino acid position is conserved. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
