Uncertain significance for Inborn genetic diseases — the classification assigned by Ambry Genetics to NM_032638.5(GATA2):c.1423A>T (p.Met475Leu), citing Ambry Variant Classification Scheme 2023: The p.M475L variant (also known as c.1423A>T), located in coding exon 5 of the GATA2 gene, results from an A to T substitution at nucleotide position 1423. The methionine at codon 475 is replaced by leucine, an amino acid with highly similar properties. This amino acid position is well conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Based on the available evidence, the clinical significance of this variant remains unclear.
