Uncertain significance for Inborn genetic diseases — the classification assigned by Ambry Genetics to NM_001283009.2(RTEL1):c.595A>G (p.Lys199Glu), citing Ambry Variant Classification Scheme 2023: The p.K199E variant (also known as c.595A>G), located in coding exon 6 of the RTEL1 gene, results from an A to G substitution at nucleotide position 595. The lysine at codon 199 is replaced by glutamic acid, an amino acid with similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
