NM_177438.3(DICER1):c.4469G>A (p.Gly1490Asp) was classified as Uncertain significance for Hereditary cancer-predisposing syndrome by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.G1490D variant (also known as c.4469G>A), located in coding exon 22 of the DICER1 gene, results from a G to A substitution at nucleotide position 4469. The glycine at codon 1490 is replaced by aspartic acid, an amino acid with similar properties. This amino acid position is well conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
