A similar copy number variant has been observed in individual(s) with prostate cancer and breast cancer (PMID: 15863663, 20617377, 20736950). This variant disrupts the FANCD2 and DNA binding domains of the BRCA2 protein, which are necessary for homologous recombination repair activity (PMID: 24323938, 12915460, 15115758, 18212739, 20450923). While functional studies have not been performed to directly test the effect of this variant on BRCA2 protein function, this suggests that disruption of this region of the protein is causative of disease. This variant disrupts a region of the BRCA2 protein in which other variant(s) (p.Leu2510Pro) have been determined to be pathogenic (PMID: 14670928, 21719596, 23108138, 29394989).
