NC_000015.10:g.(?_55234762)_(55234954_?)del was classified as Pathogenic for Griscelli syndrome type 2 by Labcorp Genetics (formerly Invitae), Labcorp, citing Invitae Variant Classification Sherloc (09022015): This variant is a gross deletion of the genomic region encompassing exon 2 of the RAB27A gene, which includes the initiator codon. The 5' end of this event is unknown as it extends beyond the assayed region for this gene and therefore may encompass additional genes. The 3' boundary is likely confined to intron 2 of the RAB27A gene. This is expected to result in an absent or disrupted protein product. For these reasons, this variant has been classified as Pathogenic. Loss-of-function variants in RAB27A are known to be pathogenic (PMID: 10835631, 23160464). This variant has not been reported in the literature in individuals with RAB27A-related disease.
