NM_006440.5(TXNRD2):c.238T>C (p.Trp80Arg) was classified as Uncertain significance for Primary dilated cardiomyopathy by Labcorp Genetics (formerly Invitae), Labcorp, citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces tryptophan with arginine at codon 80 of the TXNRD2 protein (p.Trp80Arg). The tryptophan residue is highly conserved and there is a moderate physicochemical difference between tryptophan and arginine. This variant is present in population databases (rs762714876, ExAC 0.003%). This variant has not been reported in the literature in individuals with TXNRD2-related disease. Algorithms developed to predict the effect of missense changes on protein structure and function are either unavailable or do not agree on the potential impact of this missense change (SIFT: "Deleterious"; PolyPhen-2: "Probably Damaging"; Align-GVGD: "Class C0").
