Uncertain significance for Inborn genetic diseases — the classification assigned by Ambry Genetics to NM_001211.6(BUB1B):c.1454A>C (p.Glu485Ala), citing Ambry Variant Classification Scheme 2023: The p.E485A variant (also known as c.1454A>C), located in coding exon 11 of the BUB1B gene, results from an A to C substitution at nucleotide position 1454. The glutamic acid at codon 485 is replaced by alanine, an amino acid with dissimilar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
