Uncertain significance for Spastic paraplegia — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_024306.5(FA2H):c.718A>G (p.Met240Val), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces methionine with valine at codon 240 of the FA2H protein (p.Met240Val). The methionine residue is moderately conserved and there is a small physicochemical difference between methionine and valine. This variant is present in population databases (rs367933620, ExAC 0.002%). This variant has not been reported in the literature in individuals with FA2H-related disease. Algorithms developed to predict the effect of missense changes on protein structure and function (SIFT, PolyPhen-2, Align-GVGD) all suggest that this variant is likely to be tolerated, but these predictions have not been confirmed by published functional studies and their clinical significance is uncertain.
