NM_000051.4(ATM):c.6341C>A (p.Ser2114Tyr) was classified as Uncertain significance for Hereditary cancer-predisposing syndrome by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.S2114Y variant (also known as c.6341C>A), located in coding exon 42 of the ATM gene, results from a C to A substitution at nucleotide position 6341. The serine at codon 2114 is replaced by tyrosine, an amino acid with dissimilar properties. This amino acid position is not well conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
