It is expected to result in an absent or disrupted protein product. This variant is not present in population databases (ExAC no frequency). This variant has not been reported in the literature in individuals with NF1-related disease. A different variant (c.5708delT) giving rise to the same protein effect observed here (p.Leu1903*) has been reported in an individuals with diagnosis or clinical suspicion of neurofibromatosis type 1 (PMID: 18546366, 24789688), indicating that this residue may be critical for protein function.
