Uncertain significance for Symmetrical dyschromatosis of extremities; Aicardi-Goutieres syndrome 6 — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_001111.5(ADAR):c.1313A>G (p.Tyr438Cys), citing Invitae Variant Classification Sherloc (09022015). This variant lies in the ADAR gene (transcript NM_001111.5) at coding-DNA position 1313, where A is replaced by G; at the protein level this means replaces tyrosine at residue 438 with cysteine — a missense variant. Submitter rationale: This sequence change replaces tyrosine with cysteine at codon 438 of the ADAR protein (p.Tyr438Cys). The tyrosine residue is weakly conserved and there is a large physicochemical difference between tyrosine and cysteine. This variant is not present in population databases (ExAC no frequency). This variant has not been reported in the literature in individuals affected with ADAR-related conditions. Algorithms developed to predict the effect of missense changes on protein structure and function (SIFT, PolyPhen-2, Align-GVGD) all suggest that this variant is likely to be tolerated. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
