This variant lies in the JAK3 gene (transcript NM_000215.4) at coding-DNA position 653, where G is replaced by T; at the protein level this means replaces arginine at residue 218 with leucine — a missense variant. Submitter rationale: This sequence change replaces arginine with leucine at codon 218 of the JAK3 protein (p.Arg218Leu). The arginine residue is weakly conserved and there is a moderate physicochemical difference between arginine and leucine. This variant is not present in population databases (ExAC no frequency). This variant has not been reported in the literature in individuals with JAK3-related disease.
