Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_006231.4(POLE):c.5345A>G (p.Tyr1782Cys), citing Ambry Variant Classification Scheme 2023: The p.Y1782C variant (also known as c.5345A>G), located in coding exon 39 of the POLE gene, results from an A to G substitution at nucleotide position 5345. The tyrosine at codon 1782 is replaced by cysteine, an amino acid with highly dissimilar properties. This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
