This variant lies in the RB1 gene (transcript NM_000321.3) at coding-DNA position 1815, where G is replaced by A; at the protein level this means replaces methionine at residue 605 with isoleucine — a missense variant. Submitter rationale: This missense variant replaces methionine with isoleucine at codon 605 of the RB1 protein. Computational prediction suggests that this variant may not impact protein structure and function (internally defined REVEL score threshold <= 0.5, PMID: 27666373). Splice site prediction tools suggest that this variant may impact RNA splicing. To our knowledge, RNA or protein functional studies have not been reported for this variant. This variant has not been reported in individuals affected with hereditary cancer in the literature. This variant has been identified in 1/251092 chromosomes in the general population by the Genome Aggregation Database (gnomAD). The available evidence is insufficient to determine the role of this variant in disease conclusively. Therefore, this variant is classified as a Variant of Uncertain Significance.
