Uncertain significance for Dyskeratosis congenita — the classification assigned by Ambry Genetics to NM_198253.3(TERT):c.202G>T (p.Ala68Ser), citing Ambry Variant Classification Scheme 2023: The p.A68S variant (also known as c.202G>T), located in coding exon 1 of the TERT gene, results from a G to T substitution at nucleotide position 202. The alanine at codon 68 is replaced by serine, an amino acid with similar properties. This amino acid position is not well conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
