Uncertain significance for Myopathy with tubular aggregates; Immune dysfunction with T-cell inactivation due to calcium entry defect 2; Stormorken syndrome — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_001382567.1(STIM1):c.1838C>T (p.Ala613Val), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces alanine with valine at codon 582 of the STIM1 protein (p.Ala582Val). The alanine residue is weakly conserved and there is a small physicochemical difference between alanine and valine. This variant is not present in population databases (ExAC no frequency). This variant has not been reported in the literature in individuals with STIM1-related disease. Algorithms developed to predict the effect of missense changes on protein structure and function (SIFT, PolyPhen-2, Align-GVGD) all suggest that this variant is likely to be tolerated, but these predictions have not been confirmed by published functional studies and their clinical significance is uncertain. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
