Uncertain significance — the classification assigned by Women's Health and Genetics/Laboratory Corporation of America, LabCorp to NM_001377265.1(MAPT):c.220+2503C>T, citing LabCorp Variant Classification Summary - May 2015: Variant summary: MAPT c.284C>T (p.Thr95Met) results in a non-conservative amino acid change in the encoded protein sequence. Four of five in-silico tools predict a benign effect of the variant on protein function. The variant allele was found at a frequency of 5.5e-05 in 238060 control chromosomes (gnomAD). c.284C>T has been reported in the literature in an individual affected with ALS as well as unaffected controls. This report does not provide unequivocal conclusions about association of the variant with MAPT-Related Disorders. To our knowledge, no experimental evidence demonstrating an impact on protein function has been reported.
