Uncertain significance for Glutaric aciduria, type 1 — the classification assigned by Neuberg Centre For Genomic Medicine, NCGM to NM_000159.4(GCDH):c.782C>T (p.Thr261Ile), citing ACMG Guidelines, 2015: The observed missense c.782C>T(p.Thr261Ile) variant in GCDH gene has not been reported previously as a pathogenic variant nor as a benign variant, to our knowledge. This variant is present with an allele frequency of 0.001% in gnomAD Exomes database. This variant has been reported to the ClinVar database as Uncertain significance (multiple submissions). Multiple lines of computational evidence (Polyphen - probably damaging , SIFT - damaging and MutationTaster - disease causing) predict a damaging effect on protein structure and function for this variant. The reference amino acid is predicted as conserved by GERP++ and PhyloP across 100 vertebrates. The amino acid Thr at position 261 is changed to a Ile changing protein sequence and it might alter its composition and physico-chemical properties. For these reasons, this variant has been classified as Uncertain Significance (VUS).
