Pathogenic for Developmental and epileptic encephalopathy, 9 — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_001184880.2(PCDH19):c.2617-1G>A, citing Invitae Variant Classification Sherloc (09022015). This variant lies in the PCDH19 gene (transcript NM_001184880.2) at the canonical splice acceptor site of the intron immediately before coding-DNA position 2617, where G is replaced by A; at the protein level this means a change at this position may disrupt normal splicing. Submitter rationale: This sequence change affects an acceptor splice site in intron 3 of the PCDH19 gene. It is expected to disrupt RNA splicing and likely results in an absent or disrupted protein product. For these reasons, this variant has been classified as Pathogenic. Donor and acceptor splice site variants typically lead to a loss of protein function (PMID: 16199547), and loss-of-function variants in PCDH19 are known to be pathogenic (PMID: 21053371). This variant has been observed to be de novo in an individual affected with epilepsy (PMID: 20713952). This variant is not present in population databases (ExAC no frequency).
