NM_000203.5(IDUA):c.932C>T (p.Pro311Leu) was classified as Uncertain significance for Mucopolysaccharidosis by Johns Hopkins Genomics, Johns Hopkins University, citing ACMG Guidelines, 2015. This variant lies in the IDUA gene (transcript NM_000203.5) at coding-DNA position 932, where C is replaced by T; at the protein level this means replaces proline at residue 311 with leucine — a missense variant. Submitter rationale: This IDUA missense variant (rs377684568) is rare (<0.1%) in a large population dataset (gnomAD v4.0.0: 469/1568078 total alleles; 0.03%; no homozygotes). It has been reported in ClinVar (Variation ID 648087), but has not been reported in the literature, to our knowledge. Two bioinformatic tools queried predict that this substitution would be tolerated. The proline residue at this position is evolutionarily conserved across some of the species assessed, while several have a different amino acid including a few species that have leucine. We consider the clinical significance of c.932C>T in IDUA to be uncertain at this time.
