NM_177438.3(DICER1):c.3698A>G (p.Asp1233Gly) was classified as Uncertain significance for Hereditary cancer-predisposing syndrome by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.D1233G variant (also known as c.3698A>G), located in coding exon 20 of the DICER1 gene, results from an A to G substitution at nucleotide position 3698. The aspartic acid at codon 1233 is replaced by glycine, an amino acid with similar properties. This amino acid position is not well conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
