Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_177438.3(DICER1):c.5471G>A (p.Gly1824Glu), citing Ambry Variant Classification Scheme 2023: The p.G1824E variant (also known as c.5471G>A), located in coding exon 24 of the DICER1 gene, results from a G to A substitution at nucleotide position 5471. The glycine at codon 1824 is replaced by glutamic acid, an amino acid with similar properties. This amino acid position is well conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
