Uncertain significance for Familial thoracic aortic aneurysm and aortic dissection — the classification assigned by Ambry Genetics to NM_000071.3(CBS):c.925G>A (p.Asp309Asn), citing Ambry Variant Classification Scheme 2023: The p.D309N variant (also known as c.925G>A), located in coding exon 8 of the CBS gene, results from a G to A substitution at nucleotide position 925. The aspartic acid at codon 309 is replaced by asparagine, an amino acid with highly similar properties. This amino acid position is conserved. In addition, the in silico prediction for this alteration is inconclusive. Based on the available evidence, the clinical significance of this variant remains unclear.
