Uncertain significance for Ehlers-Danlos syndrome, kyphoscoliotic type 1 — the classification assigned by ARUP Laboratories, Molecular Genetics and Genomics, ARUP Laboratories to NM_000302.4(PLOD1):c.2099A>G (p.His700Arg), citing ARUP Molecular Germline Variant Investigation Process: The PLOD1 c.2099A>G; p.His700Arg variant (rs773756799), to our knowledge, has not been reported in the medical literature or gene specific databases. This variant is found in the general population with an allele frequency in non-Finnish Europeans of 0.0018% (2/113,558 alleles) in the Genome Aggregation Database. The histidine at codon 700 is highly conserved (Alamut v.2.11) and computational analyses (SIFT, PolyPhen-2) predict that this variant is deleterious. However, based on the available information, the clinical significance of this variant is uncertain.
