NM_017534.6(MYH2):c.1228G>A (p.Gly410Ser) was classified as Uncertain significance for Myopathy, proximal, and ophthalmoplegia by Labcorp Genetics (formerly Invitae), Labcorp, citing Invitae Variant Classification Sherloc (09022015): Algorithms developed to predict the effect of missense changes on protein structure and function (SIFT, PolyPhen-2, Align-GVGD) all suggest that this variant is likely to be disruptive. ClinVar contains an entry for this variant (Variation ID: 647156). This variant has not been reported in the literature in individuals affected with MYH2-related conditions. This variant is present in population databases (rs146229690, gnomAD 0.03%). This sequence change replaces glycine, which is neutral and non-polar, with serine, which is neutral and polar, at codon 410 of the MYH2 protein (p.Gly410Ser). In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
