Pathogenic — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NC_000015.10:g.(?_55430664)_(55432608_?)del, citing Invitae Variant Classification Sherloc (09022015): This variant is a gross deletion of the genomic region encompassing exons 9-10 of the DYX1C1 gene. The 5' boundary is likely confined to intron 8. The 3' end of this event is unknown as it extends through the termination codon beyond the assayed region for this gene and may encompass additional genes. While this deletion is not anticipated to result in nonsense mediated decay, it is expected to create a truncated protein product or disrupt mRNA translation. Similar deletions of exons 9-10 have been observed in the homozygous state or with a pathogenic variant (deletion of exon 7) in DYX1C1 in individuals affected with primary ciliary dyskinesia (Invitae). For these reasons, this variant has been classified as Pathogenic.
