Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_000179.3(MSH6):c.2513C>T (p.Pro838Leu), citing Ambry Variant Classification Scheme 2023: The p.P838L variant (also known as c.2513C>T), located in coding exon 4 of the MSH6 gene, results from a C to T substitution at nucleotide position 2513. The proline at codon 838 is replaced by leucine, an amino acid with similar properties. This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis.
