NM_001999.4(FBN2):c.7007G>A (p.Cys2336Tyr) was classified as Uncertain significance for Familial thoracic aortic aneurysm and aortic dissection by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.C2336Y variant (also known as c.7007G>A), located in coding exon 55 of the FBN2 gene, results from a G to A substitution at nucleotide position 7007. The cysteine at codon 2336 is replaced by tyrosine, an amino acid with highly dissimilar properties. This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
