This variant lies in the SDHA gene (transcript NM_004168.4) at coding-DNA position 1880, duplicating one base; at the protein level this means shifts the reading frame starting at serine residue 628, producing a truncated or aberrant protein — a frameshift variant. Submitter rationale: This sequence change results in a premature translational stop signal in the SDHA gene (p.Ser628Valfs*15). While this is not anticipated to result in nonsense mediated decay, it is expected to disrupt the last 37 amino acids of the SDHA protein. This variant is not present in population databases (ExAC no frequency). This variant has not been reported in the literature in individuals with SDHA-related disease. Experimental studies and prediction algorithms are not available for this variant, and the functional significance of the affected amino acids is currently unknown.
