Uncertain significance for Hypertrophic cardiomyopathy — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_000256.3(MYBPC3):c.3773T>C (p.Leu1258Ser), citing Invitae Variant Classification Sherloc (09022015). This variant lies in the MYBPC3 gene (transcript NM_000256.3) at coding-DNA position 3773, where T is replaced by C; at the protein level this means replaces leucine at residue 1258 with serine — a missense variant. Submitter rationale: This variant has not been reported in the literature in individuals with MYBPC3-related disease. This sequence change replaces leucine with serine at codon 1258 of the MYBPC3 protein (p.Leu1258Ser). The leucine residue is moderately conserved and there is a large physicochemical difference between leucine and serine. This variant is not present in population databases (ExAC no frequency). Algorithms developed to predict the effect of missense changes on protein structure and function (SIFT, PolyPhen-2, Align-GVGD) all suggest that this variant is likely to be tolerated, but these predictions have not been confirmed by published functional studies and their clinical significance is uncertain. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
