Likely pathogenic for Primary ciliary dyskinesia — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_012144.4(DNAI1):c.1163G>A (p.Cys388Tyr), citing Invitae Variant Classification Sherloc (09022015): This missense change has been observed in individual(s) with primary ciliary dyskinesia (PMID: 21143860, 28939216). In at least one individual the data is consistent with being in trans (on the opposite chromosome) from a pathogenic variant. It has also been observed to segregate with disease in related individuals. In summary, the currently available evidence indicates that the variant is pathogenic, but additional data are needed to prove that conclusively. Therefore, this variant has been classified as Likely Pathogenic. Advanced modeling of protein sequence and biophysical properties (such as structural, functional, and spatial information, amino acid conservation, physicochemical variation, residue mobility, and thermodynamic stability) has been performed at Invitae for this missense variant, however the output from this modeling did not meet the statistical confidence thresholds required to predict the impact of this variant on DNAI1 protein function. ClinVar contains an entry for this variant (Variation ID: 646518). This variant is present in population databases (rs767667443, gnomAD 0.002%). This sequence change replaces cysteine, which is neutral and slightly polar, with tyrosine, which is neutral and polar, at codon 388 of the DNAI1 protein (p.Cys388Tyr).
