This variant lies in the AP5Z1 gene (transcript NM_014855.3) at coding-DNA position 1910, where G is replaced by A; at the protein level this means replaces cysteine at residue 637 with tyrosine — a missense variant. Submitter rationale: This sequence change replaces cysteine with tyrosine at codon 637 of the AP5Z1 protein (p.Cys637Tyr). The cysteine residue is weakly conserved and there is a large physicochemical difference between cysteine and tyrosine. This variant is present in population databases (rs557706442, ExAC 0.05%).
